I can verify that John's instructions work on my system:
>>> Gio.MountMountFlags.NONE
<enum G_MOUNT_MOUNT_NONE of type GMountMountFlags>
It looks like you are running Linux, if possible what distro?
For instance, I am running Ubuntu and the package containing gir bindings
for gio is called
gir1.0-glib-2.0 (0.9.3-0)
the relevant file containing gio bindings is:
/usr/lib/girepository-1.0/Gio-2.0.typelib
It sounds like you may have an out-of-date typelib or something?
